Higher-order chromatin remodeling promotes DNA repair by segregating damaged chromatin. Aberrant chromatin remodeling leads to inefficient DNA repair and increases tumor cell sensitivity to DNA damage. Therefore, in tumors harboring inactivating mutations in chromatin remodelers, combination therapy with PARP inhibitors can achieve synthetic lethality.
