📣 New paper online! 📣 With a team from @IKNL @radboudumc and @QueensKingston we assessed primary end point changes in #Immunotherapy trials. We advocate for more transparency. #ASCO23 #ASCO2023 #oncology
Changes to primary endpoints during or even after recruitment were omnipresent in phase-III trials with checkpoint inhibitors for urological and lung cancer patients. These changes were often not reported, as stipulated by CONSORT reporting guidelines. ja.ma/3qv10SK

A new study shows that primary endpoints are often changed after the fact in randomized trials of immunotherapy. In some cases, these changes led to substantially altered conclusions.

No matter what you were taught, don't bother to test for proportional hazards in survival analyses. Hazards are never proportional because hazard ratios vary over the follow-up if treatment has an effect on survival. @mats_julius and I explain it here: jamanetwork.com/journals/jam…

Ontluisterend fragment.
Clinical trial Q for #StatsTwitter #MedTwitter #EpiTwitter: Can you assume exchangeability between arms within strata of a patient-level factor if randomization was not stratified for this factor? And if it’s stratified for other factors associated with this subgroup factor?

“Cisplatin is preferred over carboplatin for treatment of metastatic urothelial carcinoma” Many mUC papers include such a statement and it has been in the #EAU guidelines for the last 14 years. @Uroweb But how well do the data really support it? Our new paper is about that 👇1/9

8/9 A non-randomized comparison of cisplatin and carboplatin regimens was recently performed in a Dutch cohort of contemporary mUC patients, confirming this finding as well. #bladdercancer xitter.cf/EurUrolFocus/status/14…

9/9 At the same time, higher tox levels & lower eligibility for cisplatin are well-established. In light of these data, we think revision of guideline language on this topic is necessary. Read the full OA paper here: sciencedirect.com/science/ar… @uroweb @MariaJRibal @myesmo #EAU
