8/ In summary, we have found that non-response reflects an inflammatory cytokine network that anti-TNF therapy leaves intact. The distinguishing feature is network wiring, not any single cytokine. Work done with @KorcsmarosLab @ImperialMDR. Preprint: biorxiv.org/content/10.64898…

1/ Anti-TNF agents are first-line therapy in Crohn's disease, yet many patients do not achieve remission, and the basis for non-response remains poorly understood. Our new study proposes that the cause is not a single cytokine, but the wiring of the cytokine network.

1/ Our new review in DNA and Cell Biology. Blocking one cytokine at a time rarely gives durable remission in UC. Here we make the case for treating cytokines as networks, rewired between UC phenotypes, and worth mapping in space and over time.

Very happy to have this new paper out on #Salmonella / bacterial transcription factor activity analysis. A great extension to our SalmoNet2 database, and the final paper from our projects done btw 2014-2021 in the Earlham & Quadram Institutes.
Well done @martonolbei!
Excited to share our latest extension to the SalmoNet2 database. SalmonAct is a new prior knowledge resource for inferring transcription factor activity in Salmonella. If you work on Salmonella (or bacterial transcriptomics more broadly), give it a look!

Summary of our recent publication on cytokine networks in ulcerative colitis
KorcsmarosLab retweeted
A study identifies the regulatory cytokine TL1A as an upstream regulator of the inflammatory cytokines TNF and IL-23A in ulcerative colitis and uncovers other previously unknown cytokine-cytokine interactions in the disorder. @martonolbei @TheQuadram scim.ag/46AGOkq
This is very special, not everyday you can see your paper highlighted by Science :)
Through systems immunology analysis, a team has identified potential therapeutic targets for the treatment of #UlcerativeColitis, an inflammatory bowel disease characterized by dysregulated cytokine signaling in the GI tract. @SciSignal scim.ag/4kmEVNZ

This week’s new issue of #ScienceSignaling has arrived!
A study reveals how fetal cells could activate maternal natural killer cells during early pregnancy, systems immunology reveals cytokine targets in ulcerative colitis, and more. scim.ag/4ko4K0k

To validate our predictions, we used patient-derived colonic organoids and organoid-ILC cocultures to test whether predicted cytokine-cytokine interactions actually occur in a physiologically relevant system.
KorcsmarosLab retweeted
We used systems immunology to analyse single-cell RNA-seq data from colonic biopsies of treatment-naive patients, treatment-exposed patients, and healthy donors, building cytokine interaction networks across disease states.

Why do many UC patients struggle despite cutting-edge biologics? We suspected the answer lies not in individual cytokines, but in how they communicate as an interconnected network during disease and treatment. New @KorcsmarosLab paper in @SciSignaling science.org/doi/10.1126/scis…
