Check out our recent publication in collaboration with the Khadra Lab at @mcgillu - where computational modelling 🖥️ and experimental immunology 🔬join forces to study the role of T cell diversity and antigen binding strength on chronic infection clearance! Find out more 👇
>90% of #TCR sequence diversity is generated by TdT-mediated nucleotide addition, but what is the benefit of TdT-altered TCRs? @MandlLab show that low-affinity #Tcells generated by TdT are important for curtailing chronic pathogen replication #PLOSBiology plos.io/3HJKOCl
Our PhD student Angela @ImmuneVet co-host of the TWiV podcast with @profvrr recently interviewed @bjesseshapiro and Corinne Maurice with their students Sana Naderi and Anshul Sinha about their cool virus research going on at @mcgillu! @McGillMed 📺bit.ly/3Hb9Aet
And this work featured on the current issue cover! Amazing collaboration with @JohannesTextor @MelicharLab, and the Westermann team
Wondering what we can learn from the T cell receptor beta sequence about the self-reactivity of a T cell? check out our paper in Cell Systems where we used machine learning to ask this very question👇: cell.com/cell-systems/fullte…
So proud! 🎉🌈

We add to the growing appreciation of neutrophils as a heterogenous population by characterizing differences in nuclear lobulation with respect to migratory behaviour 🌈
Our study provides evidence supporting the theory that nuclear segmentation facilitates neutrophil navigation of tight spaces, assisting their role as first-responders 🚒👨🚒
In our second approach, we isolate all the neutrophils and annotate the number of lobes in each. We track these neutrophils over time and also observe that greater lobularity correlated with greater cell speed and displacement across the channels.
Across donors, we find that the segmented and hypersegmented neutrophils more readily cross the restrictive 2 μm section compared to their banded counterparts.
We test the hypothesis in a two-pronged approach. First, we sorted the neutrophils into 3 groups enriched for banded, segmented, and hypersegmented nuclei. We differentially stained these neutrophils and imaged them by live confocal microscopy.
With @Vrisekoop team at @UMCUtrecht, we devised a system to leverage the heterogeneity in neutrophil nuclei arising from the administration of endotoxin in human donors. Neutrophils released into circulation in this model have nuclei ranging from banded to hypersegmented.
Why do neutrophils have multi-lobed nuclei? 🧐
Does this enable their rapid migration through tight spaces? 🏃♀️💨
In this study, we sought to answer this question experimentally 🔬
Here is what we found 👇
Connie Shen @peepingcells, @MandlLab @mcgillu, @Vrisekoop @UMCUtrecht and colleagues discover that nuclear segmentation allows neutrophils to migrate faster in confined spaces.

Thank you so much @CdnImmunol for the shout-out 😍! It's such a privilege to be highlighted along so many other talented scientists during Women's History Month. Here's to another year of women-led science 🥳👩🔬
Dr. Judith Mandl @MandlLab is an associate professor at McGill University and is a CRC in Immune Cell Dynamics. Her lab investigates how immune cell migration, cell-cell interactions, and naïve T cell heterogeneity orchestrate the tightly choreographed dance of an immune response
