In a separate study using experiments & continuum model, we show that cell stiffness dependent height variations are important for generating the pulsations in monolayers & in setting their lengthscales (doi.org/10.1140/epje/s10189-…). 7/7
We used a vertex model to understand the transition to pulsations & flows. We found that a dominating velocity-polarity alignment led to flows but when the velocity-polarity alignment matched the rotational diffusion, pulsations emerged. 6/7
Thus by tuning external substrate friction & the internal cytoskeletal organization we can force the monolayers to behave in a unique way - pulsate/flow. We also found correlated myosin accumulation & lamellipodial alignment during pulsations & flows, respectively. 5/7
As a standard, we then checked the role of cytoskeleton using inhibitors. The inhibitors suppressed the pulsations, but when we tried to get back the pulsations by washing out & restoring cytoskeletal activity, there were no pulsations – but long range flows. 4/7
Using spatially periodic micropatterns of various sizes, we trap the pulsations & demonstrate that substrate friction regulates pulsation sizes. By increasing the relative friction between adhesive patterns & PEG regions, we further substantiate the role of friction. 3/7
We observed the dynamics of large confluent MDCK monolayers & found spontaneously emerging, locally oscillating & flowing domains (10-15 cells ~ 200 µm). Characterizing them using divergence & velocity magnitude, we asked what determines their lengthscales. 2/7
Our study from @Raghavan_Thiaga came out recently in @iScience_CP (doi.org/10.1016/j.isci.2022.……). We show how two coexisting collective epithelial dynamics - pulsations & flows - are regulated. 1/7
