We present long-term stabilization of an oblique plane microscope (OPM). By stabilizing the remote focus unit of an OPM, we can enable long-term acquisition without light-sheet drift. The feedback signal comes from a laser, no fluorescence is expended. biorxiv.org/content/10.1101/…

Correction from @TanFad! It's actually the The Center for Advanced Light Microscopy (CALM) @UCSF that is building the #Snouty microscope! They have a great team there with @nico_stuurman, @KariHerrington and @micaelalasser (Instagram). A great location!😀

PSA: Heat-set inserts make stronger threads in a 3D print, and look quite tidy.

Following a poke from @RunningPhoton, I've fixed a bug in my KL divergence calculation function which means our parameter-free deconvolution approach now *really* has no hand-tuned parameters at all: beryl.mrc-lmb.cam.ac.uk/rlgc… Feedback always welcome. xitter.cf/JamesDManton/status/18…
We (@AndrewGYork, @RunningPhoton, @microRussell and I) are still working on the manuscript describing our automatic parameter-free deconvolution approach, but we've made a Google Colab notebook where you can try it out without needing to install anything: beryl.mrc-lmb.cam.ac.uk/rlgc…

Did you know that nuclear pore complexes can turn up outside the nuclear envelope? Studying these, @JennySachweh, Bernhard Hampoelz and others uncover that the surrounding cellular milieu determines NPC composition via the nuclear transport system itself. #teamtomo #NPC A 🧵

Where is the optical axis? Finding it can be critical to building microscopes. Here's the back reflections at ~15cm from a collimated ~1mm⌀ 532nm laser aligned into the back of Nikon 40x0.95 air objective. Radially symmetric/concentric interference rings indicate good alignment.

Thermal expansion -> a typical problem in time-lapse microscopy at high resolution. Here I measure the axial shift of the focal plane of a Nikon 40x0.95 air objective at ~1.6um/K, or about ~1.5x the depth of field per degree C: github.com/amsikking/Nikon_4…

Long awaited (thanks @JamesDManton for posting the patent application!) and now out in @ScienceMagazine: Using absorption to look deeper by @HongNeuroTech - they achieved approx. 3 mm depth using the food dye tartrazine: #Microscopy science.org/doi/10.1126/scie…

I've now made a few small changes to our parameter-free deconvolution code to better handle low SNR data, and to add MIPs to the notebook so you have a feeling for the results without needing to download images from Google Drive. xitter.cf/JamesDManton/status/18…

We discovered a new species: Stentor stipatus! We named it for its brown aggregates💩 We explore its morphology, phylogeny, and behavior in my new preprint with Daniel Cortes and @WallaceUcsf. This single cell exhibits habituation and phototaxis! bit.ly/4fwSasv 🧵1/n
