Surprise 1: Anthropic acquired a one-person biotech . . . Surprise 2: that person is me Surprise 3: I’m here to tell you all about it Yes, this is what you think it is; after 6 yrs of increasingly loose anonymity: the Professor Oak reveal post …and yes, it has gotten so boring around here that I’m literally doxxing myself to spice up the timeline ------- Ok, allow me to introduce myself. My name is Cody Tranbarger. I mostly tweet, but occasionally find time to dabble in drug dev & starting/building biotechs. My past lives span the VC, crossover, startup continuum; Prof Oak has been my way of keeping a digital foot in those worlds, despite having my physical feet planted elsewhere. The past 18 mo, ‘elsewhere’ was Mithril Medicine, a co. I founded to develop bsAbs for LAMA2-CMD & other ECM-related genetic diseases. Mithril was born out of hands-on building with @MikeNGladstone, @JasonPaulRhodes, @LifeSciVC, & team @atlasventure. Somehow, I made enough half-decent calls along the way (even a broken clock is right twice a day, eh?) to convince them Mithril/I warranted a small seed. While I can hardly rationalize such an outrageously cavalier bet, I’m grateful for it, & I'd partner with Atlas again in a heartbeat. In typical AV fashion, Mithril’s primordial form was virtual, decentralized, & ultra-lean. This stage tends to be short-lived, but for Mithril, it worked surprisingly well, for surprisingly long, thanks in part to the rapid maturation of the AI tools I used liberally. Eventually, Mithril became a bit of an experiment itself - to find the upper bound of how far it could scale as a single operator + a bunch of Claude (+ a stellar bench of part-time consultants). In parallel, I started spending time with @nc_frey, @an1lam, & @CoefficientBio team; and promptly experienced, for the 1st time, AI at the frontier. Soon thereafter, Mithril stopped looking like an experiment, & started looking like a preview of an entirely new operating model. From that spark came “Pipeline-in-a-Person.” The idea immediately resonated when we shared it here back in April; and it was central to the shared vision that led to Anthropic’s acquisition of Mithril, & my joining the team. Why join Anthropic? Three reasons. (1) For Mithril, the programs continue, with more resources & infrastructure behind them, and as much Claude as I can put to work. (2) For me, the chance to start several more programs, & find out how many one person can actually run in parallel - an almost unprecedented luxury for a curious drug hunter. (3) For our mission, a path to institutionalize & scale this new way of working, then put it in the hands of all of biopharma, all over the world - which, candidly, I find the most energizing. Will AI x Bio cure diseases? Unequivocally yes, imo… but not by the route that gets the most airtime. Rather than timeline acceleration & compression, I see orchestration & parallelization as today's low-hanging fruit. Most drug hunters see a handful of go/no-go calls in a career, & spend entirely too much of the rest trading emails (my inbox can attest). Give them the bandwidth to run several programs at once, and the time to focus on scientific judgment, and I’m willing to bet they’ll make better calls. And at scale, better calls translate to more drugs, higher hit rates, broader patient impact, & likely, a disease or 10 or 100 cured. I couldn't be more excited to build toward that future @AnthropicAI. Finally, a heartfelt thank you to BioTwitter/X. The end of anonymity is bittersweet, but if anyone’s earned a proper introduction, you all have. It’s difficult to overstate how much this app has done for me: a decade of education, plenty of friends, many job opportunities, several mentors, a few investors, even a co-founder (fmr). It was the generosity of this community that inspired me to start posting 6 years ago, after just as many years lurking. I hope I’ve been able to pay it forward, in my own small way. But, to be clear, Prof Oak isn’t going anywhere. Same beat, same themes, and now, a front-row seat to drug development at the frontier. Stay tuned for dispatches from the trenches; and remember... Gotta Cure 'Em All

Daniela Amodei reveals the woman who trains Claude's character has a PhD in philosophy, not AI or ML "It's unusual, right? You would think, oh, the person that's training the character of these models must be a computer scientist or somebody who's studied ML or AI for many decades." "Amanda's technical and she technically trains the model, but I think it is very felt in Claude's character when you communicate with it that there's this kind of experience, something that's very wise, kind of the best version of the better angel of your nature trying to answer your questions." "But if you kind of poke Claude in these difficult areas where maybe different philosophical traditions might conflict, I think that's where you especially feel Amanda because you'll sort of see like, 'oh well, you might want to consider things from this point of view but you could also consider it from this point of view.'" "I think it's important because as the world begins to rely on these models more and more, you don't want to be locked into one philosophical tradition because ethics and morality are quite complicated, and different people and different cultures might have different answers about how they would like you to behave.”

US energy company execs have resigned in disgust as the Trump family is caught secretly expediting the funneling of $25 billion from a sanctioned Russian energy giant in exchange for allowing Russians to obtain a natural gas project in Alaska. According to investigations by The New York Times and Financial Times, Russia's Novatek allegedly concealed its financial involvement in the American project through a UAE-linked intermediary. Gentry Beach, a longtime Trump ally and close friend of Donald Trump Jr., signed an agreement with Novatek to pursue natural gas development in Alaska. Reuters documented the arrangement in February. Meanwhile, Vladimir Putin discussed this cooperation on the Alaska energy project with Trump's representatives, Steve Witkoff and Jared Kushner, during negotiations involving Russia and Ukraine. The controversy deepened when Polar Train LNG's CEO, Joel Riddle, raised concerns about concealed Russian financing and possible sanctions violations. He departed in September, followed by other senior executives. The Financial Times also described discussions about a potential arrangement under which Novatek would provide technology for the Alaska project in exchange for sanctions relief benefiting its Arctic LNG 2 development in Siberia. According to The New York Times, Riddle and two other departing executives warned the Trump administration about the Russian financing and urged officials to investigate. The White House maintains that the project was mentioned only briefly and that no detailed discussions took place. There is no established evidence that Kushner or Witkoff knew about the allegedly concealed financing. Polar Train LNG denies wrongdoing. The central question is how a sanctioned Russian energy company became involved in a major American energy venture while Putin was discussing potential economic cooperation with Trump's representatives and a longtime Trump family associate was pursuing a separate agreement with the same Russian company. Selling Alaska to the Russians or selling Ukraine to the Russians - to the Trump family it's all the same. ft.com/content/e9cfbfbb-e65a…

Absurd dominance from 6.1 Sol. Same cost as V4.1F, radically better results. And a hole in "V4.1F-1017 is a 5.6 Sol replacement" (though this was known in this case, 5.6 was already maxing out the soft ceiling of the original CritPt) Grok 4.7, on the other hand, surges to parity
After 4 months of careful expert review, we release CritPt v2. GPT-6 Astra leads the way with 83.1% accuracy (average of 5). We made changes to 47 challenges in total, which breaks down to: - 5 corrections to mistakes in the problem statement (e.g., wrong number, self-contradictory constraints) - 19 revisions to address underspecification (e.g., missing conventions/constraints that gives ambiguity to final answers) - 16 grader fixes (e.g., grader unstable, error tolerant range too tight) without revision to the problem statement - 7 refinements even though no issues were found (e.g., rephrased to be less searchable, worded more rigorously) We also fixed some minor typos (spelling, grammar) which are unlikely to affect model performance. Here’s how we found the issues: - David Louapre @dlouapre from HuggingFace reported 10 issues across 8 challenges in June 2026. We accepted 8 issues and revised 8 challenges accordingly. We also thank David’s helpful revision suggestions. - Anthropic sent a list of ~65 issues across 31 challenges in Aug 2026. Upon careful review of the full list, we accepted ~28 issues and revised 19 challenges accordingly (two overlapping with David’s list). - We conducted an internal expert review of all problems, including newly revised ones, involving cross checking full responses of GPT-6 Astra (max) with CritPt experts. We ended up fixing/refining 20 additional problems. From 28.9% in Nov 2025 (GPT-5 high, coding tool) to 83.1% now (GPT-6 Astra max, coding tool), the progress AI has made on well-posed physics problems is remarkable in a year. CritPt consists of entry-level research problems for junior researchers: the kinds of warm-up exercises a hands-on advisor might pose for a fresh grad student, but still demand solid physics training and nontrivial physical reasoning. In this sense, frontier models are crossing a “critical point”, but there’s a lot more beyond this transitioning point in real scientific discovery. Asking a good question can be much harder than solving one. A good question can be fundamentally ambiguous and constantly changing as we explore. And ultimately, decisive tests of physics require experiments. There is so much more to explore. We sincerely thank our evaluation partner @ArtificialAnlys, who will co-host the leaderboard of CritPt v2 soon! 🚨 A MASSIVE internet security emergency is unfolding right now. 🚨 Two CRITICAL vulnerabilities have been discovered in PHP, potentially putting millions of websites at risk. → NEARLY 70% of websites with a known server-side language run PHP. The potential scale is ENORMOUS. → TWO CRITICAL security flaws have been confirmed, each scoring 9.2/10 in severity. → Attackers could potentially take control of vulnerable servers remotely, without authentication. → Reports suggest PHP versions dating back to 2004 could be affected. The exact scope is still unconfirmed. → EMERGENCY PATCHES are scheduled for TOMORROW at 10 AM ET. Website owners are being urged to prepare immediately. → Europe's CERT-EU is warning organizations to patch ASAP or consider TAKING EXPOSED APPLICATIONS OFFLINE if they cannot be secured. → WordPress, Magento, Laravel and potentially millions of other PHP-powered websites could be at risk. → Once patches go public, hackers could reverse-engineer them and race to attack servers that haven't updated. The vulnerabilities are CONFIRMED. The full technical details remain secret. Tomorrow, the race between hackers and defenders begins. This could get ugly. FAST.
The PHP security team is giving advance notice: Upcoming security release Wed, Oct 21st 2026, 14:00UTC, two critical vulnerabilities, both rated CVSSv4 9.2. PHP users are encouraged to prepare to update. Earlier release if details become public or active exploit evidence.

🚨 BREAKING: Node.js just dropped its OWN security bombshell. 4 HIGH-SEVERITY vulnerabilities patched TODAY, including one that could let hackers remotely execute malicious code on vulnerable servers. → 10 SECURITY FLAWS were addressed in today's Node.js release, FOUR rated HIGH severity. → REMOTE CODE EXECUTION: CVE-2026-111497 is a serious flaw in Node.js's Brotli decoder that could allow attackers to execute malicious code remotely. → TLS SECURITY BYPASS: Two separate vulnerabilities could trick affected applications into accepting certificates for the WRONG server under certain conditions. → Think about that. Your application could establish an encrypted connection while failing to properly verify WHO it's talking to. → THREE MAJOR NODE.JS VERSIONS AFFECTED: Node.js 22, 24 and 26 all required emergency security updates. → RUNNING NODE.JS 18 OR 20? Those versions are END-OF-LIFE and aren't receiving these official security patches. → PATCH IMMEDIATELY: Update to Node.js 22.23.4, 24.22.1 or 26.12.1, depending on your release line. If you're using Docker, rebuild and redeploy your containers. PHP AND Node.js both facing major security disclosures on the SAME DAY. If you run a website or application, TODAY is the day to check your servers. And remember: the public release of security patches can give attackers a roadmap to systems that haven't updated. Don't be the last one to patch.
