ALT Mitochondria in Health & Disease 1 Nov 26 In this edition -Stimulated protein degradation to promote mitochondrial function and neuronal survival (Chandrasekhar et al., Science). Activating the reductive-stress E3 ligase CUL2-FEM1B preserved mitochondrial function and neuronal survival in models of COX deficiency syndrome and Friedreich’s ataxia. -MCT1-dependent mitochondrial lactate import sustains cardiac metabolism (Cluntun et al., Nature). MCT1 sits in the inner mitochondrial membrane of cardiomyocytes and, with matrix LDHB, turns lactate into pyruvate and NADH to fuel the electron transport chain. -Respiration-deficient cells require pyruvate carboxylase to suppress asparagine auxotrophy (Cui et al., Nature Metabolism). Patient-derived thyroid tumour cells with high-heteroplasmy complex I mutation and low PC are sensitive to L-asparaginase.
