And folks, I gotta say it. @CurrentBiology is the Journal of Choice for @papersofnote. Consider this the seal of approval!
Anyway, tl;dr is that vesicles (SSVs and DCVs) and the cytoplasm only have a small pH difference in the fly NMJ. This is likely also true in central brain neurons, though not directly measured here. No no nice easy pHluorins for us. 😭
How cute of a crustacean? Parhyale hawaiensis is so adorable, has a fully sequenced genome, can be CRISPR'd, and lives in shallow water in tropical regions where you'd imagine the tides would be an important cue. So does it have circatidal rhythms, and can you make tides in lab?
I try to be nice about the papers I tweet, but I gotta say this one is doing me a confuse. The paper claims to be the first to show that some clock neurons express insulin, which it isn't. Also investigates a role for Smads in regulating insulin signaling. doi.org/10.1371/journal.pone…
There are 8 Tot genes in all, with only TotC here showing a role in heat tolerance. What do the other Tots do - some, like TotA, are induced by heat, but don't seem to affect heat tolerance. TotA does affect cold tolerance in another paper tho. I wish we knew more about the Tots.
This paper also provided me with the new derogatory term "time-ignorant competitors" for people who don't keep track of circadian time in their experiments.
cAMP imaging has really lagged behind GECIs in all systems, so I'm excited to see all the new science that a fast, sensitive, user-friendly sensor will reveal! Key question: When will these fly lines be @flystocks ???
Actually the chimera used here has 3 substitutions (W362F, S367T, V377T) conferring a 2000X increase in C-type inactivation rate. And check this out! Big ole vestibule opens up at the extracellular end of the selectivity filter!
However, it's been structurally difficult to capture a C-type inactivated channel. But they do it! ~3 Å structures of shaker (with the inactivation domain removed) and the W434F C-type inactivated channel. And tl;dr just look at that p-loop/filter conformational change!
Can we just marvel at structural biologists trying to describe proteins with words for a minute? "The shape of a figure of eight." "Grip the interface." "Floating on the periphery." "Elongated, bean-like shape." "Poised like the handles of an accordion."
This makes for a sorta weird-clock as shown in Fig. 5H wherein the negative regulator PER (maybe with a partner?) inhibits ROR and REV-ERB activation of rhythmically expressed genes, but the rhythm is larval staging, not days.
