Exciting talk from @PaternaBio. If they can mature sperm in the lab, men with severe infertility could have biological kids.
The same tech could let us genetically screen sperm before conception, making embryo screening far more powerful.
Very excited to be attending this talk from @PaternaBio founder @apastusz today!
Kudos to @nucleusgenomics for sponsoring this IRB study testing whether screening embryos based on polygenic risk scores will decrease implantation rates compared to screening only on embryo morphology!

Today we launch NeuroRisk, the first embryo test for autism and schizophrenia that looks for mutations in evolutionarily important genes.
These mutations are individually rare but over 10% of people carry at least one mutation in this class.
Hi Kian, great to see more IQ PGS research! A few things:
* You say "optimize embryo DNA for 14 IQ points". I think you're referring to the expected spread for 10 embryos, but this is a misleading way of stating these results. Expected gain is the logical number to share. (To be clear, I think that Herasight sometimes summarizes things in a similarly unhelpful way, which should also be called out.) Further, the relevant number is achievable births; 10 achievable births is quite optimistic (and/or expensive) for most people at the moment.
* You say in the context of the IQ model that "The models were trained on 1,000,000+ people", but that contradicts the 500k number from the paper; you may be referring to the data for the height and BMI models, but that doesn't match the context.
* You say "validated across 40,000+ siblings"; that's not for IQ though, right?
* You say "Parents across the world now have the choice to substantially increase their child’s intelligence."---but there are quite substantial dropoffs for many ancestry groups for PGSes trained on mainly European-ancestry data. This is a great aspiration, but it overstates the current state of things.

AI is rapidly getting smarter.
Now, humanity can too.
Today, @nucleusgenomics is announcing Vitruvian, our newest set of genetic optimization models.
Vitruvian’s intelligence model can optimize embryo DNA for 14 IQ points — nearly a standard deviation.
The models were trained on 1,000,000+ people, validated across 40,000+ siblings, and used more than 7 million genetic markers.
In Superintelligence, Nick Bostrom proposed genetic optimization as a key way for humanity to keep pace with rapidly advancing AI.
Bostrom’s vision is no longer theoretical.
Genetic optimization, like AI, has followed a scaling law: as datasets have grown, so have model capabilities. This trend will continue.
Parents across the world now have the choice to substantially increase their child’s intelligence.
And the next generation can choose to do the same.
AI is no longer the only intelligence that will compound.
Humanity can now direct its own evolution.

People are in favor of reprogenetics to decrease disease risks. Other traits are more controversial, but the main use of reprogenetics is not opposed by the public, it is widely and strongly supported.

A major source of attrition in the IVF process is missegregation of chromosomes in the oocyte. That can happen both during the completion of meiosis I in preparation for ovulation, and after fertilization when meiosis II is completed. The oocyte might fail to successfully have its spindle fibers attach to the kinetochores on the chromosomes and pull away a full set of chromosomes from the rest; this failure leaves an aneuploid set of chromosomes. This happens especially in aging oocytes, apparently in part because of fragmented kinetochores.
Two companies, Ovo and U-Ploid are developing methods to support correct segregation in oocytes. If these methods worked, IVF might be twice or more effective, resulting in many more euploid embryos on average.
Announcement of very cool research by Conception Bio, combining two decades of work from the field: They took human iPSCs through germline development, producing mini-ovarian organoids with follicles containing primary oocyte-like cells entering meiosis--all iPSC-derived. A remaining barrier to full in vitro oogenesis is progressing past the pachytene stage of meiosis I, to form crossovers and actually undergo meiosis. Another major task will be validating the epigenomic competence of in vitro-derived oocytes to form a healthy embryo.

I’m so excited to share this update on @Conception –
We’ve generated the first early human eggs derived from stem cells.
This is a big deal -- the potential to redefine fertility is real.

Actually using gene editing for reprogenetics has big obstacles. How to make many edits? How to make them precisely? Dr. Eriona Hysolli, former lead of the project at Colossal to de-extinct woolly mammoths & now stealth founder, will speak at ReproFro on her work. Tickets⬇️
