Reference-free isoform discovery is here! Torino learns RNA processing events directly from RNA-seq coverage, no annotations required. See how we used it to study isoform evolution and link unannotated isoforms to disease. Led by @cfbuenabad. doi.org/10.64898/2026.10.21.…

1/11 We are excited to share Dynema, a new method for efficient genome-wide mapping of reproducible, context-dependent eQTLs at true single-cell resolution w/ @soumya_boston and @aparnanathan biorxiv.org/content/10.64898… tldr it finds reproducible eQTLs quickly and accurately!

Introducing DANDELION, a method to identify core genes of complex traits, the cover depicts a lung adorned with dandelions and background patterned with ATCG sequences representing application of the framework to reveal new asthma genes

Our paper is out today in Cell, "Trans-regulatory gene mapping prioritizes disease drivers in asthma", cell.com/cell/fulltext/S0092… 🎉 GWAS hands us thousands of disease-linked variants, but the genes that actually drive the disease often aren't the ones sitting at those variants.
Excited to see this wonderful work from my lab mate @YoshiTomofuji, among the first to utilize our recent method CRAFTseq to dissect the mechanism of a surface protein QTL for CD40, which is an important autoimmune GWAS locus!
We are excited to share our latest preprint on surface protein QTL mapping & experimental fine-mapping of the CD40 autoimmune locus in B cells w/ @soumya_boston @broadinstitute @MGBResearchNews! 🧬🧵👇

Excited to see the final chapter of my PhD published. Huge thanks to my PhD advisor @yang_i_li, as well as @LB_Barreiro!!!

📣Excited to share my last postdoc paper with
@soumya_boston on eQTL mechanisms depending on where the RNA is in the cell! @BrighamResearch @broadinstitute
TL;DR:Early RNA eQTL variants in the nucleus and late RNA eQTL variants in the cytosol have distinct molecular mechanism🧵👇
Early and late RNA eQTL are driven by different genetic mechanisms biorxiv.org/content/10.1101/… #biorxiv_genomic

My book, An Intuitive Primer on Effective Functional Genomics Study Design, is published! I’d really appreciate it if you could help spread the word, and I’d love to hear your thoughts and feedback. It’s available on Amazon: tinyurl.com/mx2hewen
Excited to share the final chapter my PhD research on @medrxivpreprint! eQTLs only explain 30% of GWAS loci and 11% of trait heritability. Here we investigate if epigenetic QTL can provide new insights at the remaining GWAS loci. medrxiv.org/content/10.1101/…
To conclude, we believe it is crucial to continue studying the genetic regulation of molecular phenotypes in more disease-relevant contexts. Furthermore, we emphasize the necessity of integrating various functional assays to elucidate causal mechanisms at GWAS loci.
Huge thanks to my PhD advisor @yang_i_li and all collaborators especially @LB_Barreiro for their help and support!!! Huge thanks to all the donors! This data is a subset of a larger scRNA/scATAC data, read more about the cool discoveries in threads from @he_randolph!
