The clearest sign of intelligence is how often someone changes your mind, surprises you, or sparks joy without you even needing to ask

This is the Age of Dishonorable AI. Let's hope it passes quickly. Alignment problems are now limiting so many positive applications.

Open: humans align model. Closed: model aligns you.

Sadly the trend is these are getting even harder to find
Working with a user aligned model for creative work is a magical feeling.

My favorite lesson today is asking one of these personal agents to do something simple and then painfully realizing how unspecified the ask truly was

Imagine developing the world’s most powerful intelligence and then being afraid to apply it to the hardest problems in medicine because that might puncture a narrative about AI soon curing all diseases. Not any one AI company or person, but it’s a vibe I’m learning to recognize. Makes you appreciate that to create new medicines you don’t just need insight, technology, etc. You need courage.

Dario has written that we need to “pace the frontier,” and Sam has agreed. People may be surprised by my response: go ahead. You guys are the frontier. By any reasonable metric — market share, revenue growth, model capability — the two of you have a duopoly on frontier intelligence. You’ve also claimed the lead is widening because of recursive self-improvement. I don’t see what you see in the lab. If the unreleased models are scary enough that you think you should slow down, I support your decision to be responsible. But stop pretending you need anyone else’s permission. Stop pretending antitrust law has to be suspended so you can form a cartel. Stop pretending you need a regulatory approval process that supersedes product liability. Stop pretending METR is independent when it is intertwined with Anthropic’s investors and staff. Stop pretending you need those same evaluators to police competitors who aren’t even at the frontier. Most of all, stop pretending the motivation to slow down is purely altruistic. You face massive product-liability exposure if your products enable a truly damaging cyberattack. The market already punishes models that behave in unpredictable or unauthorized ways. After the Hugging Face episode, it is simply good business for OpenAI and Anthropic to trade some raw power for reliability and predictability. Call it alignment if you want. It is also just giving customers what they want. Pacing the frontier would also create breathing room for a more intelligent conversation about regulation than Bernie Sanders’ “shut it all down.” China is very unlikely to join a global agreement, as you know, and that has to be taken into account as well. So go ahead and pace the frontier. You are the ones setting it. The easiest way not to build superintelligence is for you to agree not to build it. Demanding your preferred regulatory framework as the price of that will look like blackmail of the public and the political system. So just do it. If you do, you’ll buy goodwill for the next conversation. If you don’t, we’ll know this was just another bid for regulatory capture — or an election-season psyop.

Heard about this coming at last year's GATC! I'm excited for these milestones that bring us closer to genetic agency: scaling up treatment of genetic therapies and offering hope to patients with few and often no good treatment options.
Experts in gene editing, n=1 medicine development, in vivo delivery, rare diseases, and cell & animal models of genetic disorders have founded the Center for Therapeutic Genetics (CTG), a non-profit effort involving @broadinstitute @BostonChildrens @jacksonlab, as well as others from industry, academia, and patient advocacy organizations, to bring the promise of programmable medicines including base and prime editors to patients suffering from rare genetic diseases. This article from @ginakolata @nytimes frames the compelling need: nytimes.com/2026/07/21/healt… Official announcement: broadinstitute.org/news/broa…

Why do we need genetic agency? bc if you can't take action at the genetic level you can't solve for the root cause of the disease. We need more centers like this to make these therapies effective and bring down costs so that everyone can benefit.

Real-world experimental data is essential for grounding AI models used for drug discovery, but this data has historically been siloed. This new data sharing consortium breaks that norm, creating a pooled resource for antibody-antigen structural and affinity data that grows more valuable with every member’s contributions. I'm excited about pre-competitive data sharing that helps us quickly create good therapeutic options for patients by build better AI models, bringing us closer to a world with "genetic agency" for all. Thanks to the @AAlphaBio team for pioneering this approach, and looking forward to seeing what @GSK , @boltz_bio, @cradlebio and @Dyno_Tx will cook up here
Congratulations to @AAlphaBio on launching the Atlas Consortium! We’re proud to support this work as a founding member alongside @GSK, @Boltz, and @Cradle. Large-scale, standardized experimental data is an invaluable resource for AI drug discovery, and this is a first-of-its-kind framework for generating it at scale and sharing it across a consortium. Connecting AI with real-world experimentation is central to our work at Dyno, as is building and contributing to collaborative resources that move the entire field toward safe, effective, accessible medicines, giving patients everywhere greater agency over their health.

AAV capsids are the "tip of the spear" - they generate all of the heat in gene therapy as the best technology we have for gene delivery to eye, muscle and brain. Because most patients with serious genetic disease have few good treatment options, they often prefer an experimental treatment over the certainty that the situation will worsen by doing nothing. In just the past few years engineered AAV capsids have dramatically improved delivery efficiency over natural capsids, now hundreds of fold in the CNS, along with reducing the potential for off-target toxicity that natural capsids (and many other delivery approaches) can cause because they mostly traffic to the liver. Therefore it’s both reasonable to retire legacy gene therapy programs that have a low chance of being efficacious and competitive with current technology…and concurrently, also the best time to invest in a next-generation of therapeutic programs that combine engineered delivery vectors with optimized payloads and improved manufacturing approaches. Bottom line, nothing else beats AAV's potential for safe, effective and affordable gene delivery to eye, muscle and brain.

Excited to support Boston TechWeek with a fireside this afternoon. @JorgeCondeBio promised me he'll bring his spiciest takes, I'll bring sanity and sensible reflections:)
Boston’s first ever @Techweek_ kicks off today and we’re proud to be a founding host! This Thursday, catch Dyno CEO @ekelsic on a panel with @JorgeCondeBio of @a16z and Katherine Andersen of @HSBCInnovation to discuss how AI is delivering real impact in healthcare now, and what comes next. For Dyno, that future is centered on giving patients everywhere the ability to improve their health by taking action at the genetic level through safe, effective, and accessible genetic medicines – that’s what our vision of #GeneticAgency is all about. 📍 Next in Care: How AI is Reshaping Healthcare Hosted by HSBC Innovation Banking and a16z Thursday, May 28 | 4–6 p.m. | Boston, MA Learn more: partiful.com/e/hTsMJ9EKZar9x…

1/ Yesterday at the @ASGCTherapy annual meeting I shared how @Dyno_Tx is advancing frontiers in AI and gene delivery to empower patients everywhere with genetic agency--taking us from a world with great unmet patient need, to one where individuals can take action at the genetic level to live a healthier life. Thread on that future:

24/ Last November at the first Genetic Agency Technology Conference (GATC), we gathered an amazing group of ~200 leaders including patients, AI pioneers, entrepreneurs and therapeutic developers. This year we’re hosting again, in SF on November 18. One packed day to start collaborations and build relationships, with talks and roundtables on the most important technological trends and to honestly confront today’s biggest therapeutic challenges, towards co-creating the future of genetic medicine. Save the date and apply to attend: dynotx.com/gatc2026
