AlphaFold is moving from structure prediction to function discovery. A new Nature Communications study introduces MitoMatch, repurposing AlphaFold-Multimer to screen 630,003 protein pairs across the human mitochondrial proteome. → 2,895 predicted interactions → partners identified for 85 uncharacterized mitochondrial proteins → orphan proteins linked to known biochemical pathways → evolutionary conservation tested across 11 eukaryotes → experiments helped define the CoQ metabolon and connect COA4 to mitochondrial copper delivery Instead of asking only: “What does this protein look like?” AI can increasingly ask: “Who does it interact with — and what does it do?” A compelling step from structure → interaction → biological function. nature.com/articles/s41467-0…

Congrats to @biozentrum @snsf_ch Ambizione Group Leader @FlorentWaltz on this stunning study🤩 of respirasome structure & organization inside native cells! It's amazing how quickly in situ #cryoET is advancing. Read Florent's thread🧵 for the high-resolution details🔎🦠 #TeamTomo
Super excited to share our latest work on the native in-cell organization of the mitochondrial respiratory chain 🥳 Using cryo-electron tomography🔬, we show how the respiratory complexes (and other complexes) are organized inside native mitochondria!  biorxiv.org/content/10.1101/…

I also would like to say that none of this would have been possible without the amazing people developing the cryo-ET software. I consider myself incredibly lucky to have been working with Ricardo Righetto in our team, he made sure all the software were properly working.

CoQ10 is a very hydrophobic molecule with poor bioavailability. In this new study, led by @LauraSteenberge, we build on prior work to show that the much more soluble CoQ4 is sufficient to rescue diverse CoQ functions and can be targeted to mitochondria. doi.org/10.1016/j.jbc.2024.1…

Mechanistic studies of redox dysfunction remain challenging due to limited strategies that can perturb redox metabolism in various cellular or organismal backgrounds. Most studies involving impaired redox metabolism have focused on oxidative stress; consequently, less is known about the settings where there is an overabundance of NADH reducing equivalents also known as reductive stress. In an article in Nature Chemical Biology @nchembio today, we are introducing a new genetically encoded tool to increase cellular NADH/NAD+ ratio which we named EcSTH. Using EcSTH, we determined that metabolic and transcriptomic signatures of the NADH reductive stress (aka high NADH/NAD+ ratio) are cellular background specific. For example, we show that, under the NADH reductive stress, non-oxidative PPP, purine biosynthesis and GDF15 metabokine levels are upregulated. Another interesting implication of our work is that reductive stress can also result in the accumulation of noncanonical sugar phosphates, which are known to be toxic. We anticipate that EcSTH tool will be useful for investigating redox metabolism in various cellular and organismal contexts and disease states, including aging-associated metabolic changes! @XingxiuPan @MinaLorrain @saraviolante @russpgoodman Check out the full version of the paper below for details: rdcu.be/dpypb or nature.com/articles/s41589-0… Supported by: @Scintillon @scrippsresearch @NIGMS @NIDDKgov
