Phase II REPRINT: Enfortumab Vedotin Plus Pembrolizumab in Rare Renal Carcinomas At the ESMO Congress 2026, Giuseppe Procopio presented preliminary first-stage results from the Phase II REPRINT trial (NCT06302569), evaluating enfortumab vedotin plus pembrolizumab in patients with collecting duct carcinoma (CDC) and renal medullary carcinoma (RMC). Key findings: ▫️ Study population: 13 patients enrolled; nine evaluable after central pathological review (seven CDC and two RCCU-MP). ▫️ Objective response rate: 22% (2/9), with both responses occurring in patients with SMARCB1-deficient RCCU-MP. No objective responses were observed in CDC (0/7). ▫️ Disease control rate: 55% (5/9), including two partial responses and three cases of stable disease. ▫️ Median PFS: 5.2 months (95% CI 3.4–not reached). ▫️ Safety: No Grade 3–4 adverse events were reported. ▫️ Biomarkers: Both responding tumours were negative for membranous NECTIN-4 expression. Takeaway: REPRINT demonstrated preliminary antitumour activity and met the prespecified criteria to proceed beyond the first stage of Simon’s two-stage design. Meet-URO 33 Study: Bone Metastases, SREs and Bone-Targeted Agents in mRCC A new subanalysis of the Meet-URO 33 study evaluated the prognostic role of bone metastases (BMs), skeletal-related events (SREs), and bone-targeting agents (BTAs) in patients with metastatic renal cell carcinoma receiving first-line systemic therapy. Among 1,696 patients, 526 (31%) had BMs at metastatic diagnosis. Median OS was 26.9 months in patients with BMs and was not estimable in those without BMs. Median PFS was 14.2 vs 19.4 months, respectively. BMs remained an independent prognostic factor for OS in multivariable analysis. Among patients with available SRE data, 26.8% experienced at least one SRE. BTAs were initiated concomitantly with first-line systemic therapy in 27.4% of patients with available BTA data. Among patients who had not experienced an SRE before systemic therapy, SREs occurred in 22.2% of those receiving BTAs versus 13.7% of those not receiving BTAs, a finding the authors noted was likely influenced by selection bias.
