I’m delighted to share our @NatureCancer paper by Anne Kraemer et al. nature.com/articles/s43018-0… Our objective was to investigate whether there are substantial differences in the antigenic landscape presented in tumors that are T cell inflamed versus those that are non-inflamed.

Great collaboration from ⁦@NickRettko⁩ and ⁦@lstops⁩ from Forest White's lab where we show MEKi can increase presentation of pMHC's derived from tumor-associated antigens, which can be targeted with BiTEs for enhanced cytotoxicity in melanoma! pnas.org/doi/10.1073/pnas.22…

We also got @lstops to measure, for the first time everrrr, the number of peptides cross-presented per cell using quantitative mass spec! This validated our basic finding that membrane-associated proteins are poorly cross-presented. Such a cool technique, I'm still in awe.

Here's an approach that will find many applications for years to come—a new mouse model enabling deep cell-specific profiling of the immunopeptidome in vivo. Amazing to see @ajaeger40's vision come to life and a privilege to be an extra in this tale. Congrats to all involved!
Proud to share my work out today in @Nature interrogating antigen presentation in vivo! We found that physiological context shapes the antigenic repertoire of cancer cells and describe a new tool to uncover tissue and cell type specific antigens in vivo. nature.com/articles/s41586-0…
